So. I was just thinking about you the other day and today as well during lab work, and I must say that I admit that I dropped agar plates with cultured E.coli because my mind was not thinking straight, that it was focused on something else aside from work. So, to who ever you are, how are you? Long time naman wa ta naka talk talk uy, sigoro kai busy man gud atong schedule, pero na happy baya ko that we were able to talk again. I understand pood busy kaayo imung schedule, don't worry na lang, it is of no concern. Gusto lang ko mo write aning letter kai ive been thinking about you and wanted to get it out of my chest. Sus, na unsa man ko uy. It feels good to write this out tho, short one.

Materials and Methods: (Rough Draft)

Study Site: Sandy Lake

The study site was Sandy Lake, a limnological site in the periphery of Stoneboro, which is in the parameters of Mercer County, Pennsylvania. The study site was the lake itself as well as the surrounding streams and man made channel that once demonstrated intervention in terms of preventing excessive metal effluents from entering the lake bed, particularly during the 19th century as a result of coal mining and industrialization. The bed of Sandy Lake was the main focus, which is where the core extracts were taken from for further CNS and metals analysis.

Materials

The materials that were used in this study were one-hundred 10-inch plastic blags, two 3-inch foot plastic cores, a coring extractor machine, a row boat with paddles, gravity core and piston, 69 aluminum plates, 69 plastic tubes, 69 glass capsules, 60 Erlenmyer flasks, a 50 ml graduated cylinder, 5 standard academic hot plates, standard titration kits, 69 ceeramic tubs, aces to the CNS machine, access to the AAS machine and about 5-10 gallonts of distilled water, 2 bottles of 30-35% H2O2, concentrated HNO3 and HCl.

Preparatory Procedure

The acquisition of core samples was conducted in Sandy Lake; two core samples were taken and properly placed in plastic tubes for properly analysis in lab. The core samples were immediately stored in the ecology refrigerator in order to prevent form gaseous reactions from occurring while preparing for analysis (Schwoerbel, 135-140). The refrigerator was a standard academic fridge; temperature was at around 30 degrees Fahrenheit.

After preparation was completed, the sediments were removed from the core tubes with a piston extruder in 1 cm thick sections and were placed in pre-labeled plastic bags according to the centimeter depth of the sediment. After the extraction of the first core, the second core was then assessed, which were also placed in the same bags as with the first core. Overall, 69 bags were used to contain the 69 mud samples. The sediments were refrigerated at 30 degrees Fahrenheit to prevent the formation of gases from occurring, while preparing for further analysis. I then prepared 69 foil pans, which were used to hold the sediments while it dried in the laboratory oven at a temperature of 50 degrees Fahrenheit. The weight of the foil pans differed and ranged from 1.00 g to 1.42 g. Prior to heating, I weighed all samples to acquire the fresh weight. After weighing all samples, 22 samples were heated on one week at 40 degrees Fahrenheit, and allowed at least 50 hours to pass for proper heating and drying of the said sediment samples. I properly heated and dried the remaining 47 mud samples, drying it at 50 degrees Fahrenheit in a 50 hour time frame. Overall, this process took about 1.5 weeks.

After the 69 samples were completely dried, they were reweighed to acquire the dry sample weight. In the meanwhile, 69 plastic tubes and covers were prepared and properly  labeled according to sediment depth. I then took all the dry samples and crushed them vis-à-vis mortar and pestle. The resulting sediment powder samples were placed in their individual plastic tube capsules for processing.

CNS Analysis

I then prepared the 69 dried samples for CNS analysis, which required me to prepare 69 ceramic tubs that were pre-washed and dried. I then prepared aobut 0.10-0.20 g of each of the powdered sediment samples for analysis, which were weighed out to at 10^-4 accuracy. The ceramic tubs, after drying, were then weighed and averaged at 36.85 g. The remaining dried and crushed sediment samples were kept and reserved for acid digestion, which is the preparatory step prior to metals analysis. All 69 samples were then  processed through the CNS machine. The elemental analyzer (EA) combusts a sample in a high temperature furnace. Carbon, nitrogen and sulfur compounds are oxidized to CO2, NOx, and Sox, respectively and these gases are separated in a gas chromatograph column then fed sequentially into an infra-red gas analyzer. After calibration with a standard known Carbon, Nitrogen, and Sulfur content, the analyzer then produces data directly as present composition. The total time it took to process the calibration samples as well as the 69 sediment samples was around 4 days.


Acid Digestion

About 0.1 g of oven dried sediment from Sandy Lake was weighed out and measured to the nearest 0.0001 g. I then quantitatively transferred the weighted dried sediment into a 125 Erlenmyer flask and added 5 mL distilled water and 5 mL concentrated HNO3. I then gently boiled the sample on a hot plate until the volume was reduced by about a half, which took about 40-50 minutes. It was rather imperative to remain in the work site to prevent excess volume from dissipating. After half of the known volume was evaporated, added 5 mL of concentrated HNO3 was added and allowed to boil until the volume was yet again reduced to half. The time it took for this to occur was about 40-50 minutes.
   
Afterwards, the resulting mixture was removed from the hot plate and 2 ml of distilled water and 3 ml of 30% H2O2 were added individually. The sample then effervesced; then added a second batch of 3 ml aliqot of H2O2. I then returned the sample back to the hot plate and allowed it to reflux, placing a watch glass in order to prevent excess evaporation. The mixture was allowed to reflux for about 2 hours. After the 2 hours passed, added 10 ml of concentrated HCl and then recovered the flask with the watch glass and allowed the samples to reflux yet again for another 15 minutes. After that, the samples were removed form the hot plate and allowed it to cool for about 15-20 minutes. After sufficient cooling time passed, transferred the liquid samples into a 50 ml graduated cylinder and then added distilled water to fill in the 50 ml graduated cylinder. This was done individually for each sample. Afterwards, placed the sample in pre-prepared glass capsules. This part of the experiment required 5 batches to be prepared and completed, which included all 69 samples. The total amount of time it took me to finish this part of the experiment was about 3 weeks.

Trace Metals Analysis

I then proceeded to take the samples to be analyzed for trace metals analysis. The key metals being studied were zinc, lead, cadmium, arsenic, and mercury. All 69 samples were taken to the AAS machine (atomic absorption spectrometer) and ran calibrated samples of lead, zinc, cadmium, mercury, and arsenic before analyzing all of the 69 samples. Each sample was analyzed for presence of the said 5 metal elements and data was processed.

As for me, Ate Ging, there is not one moment where I do not think of my baby brothers and my parents. Last time I went back home for winter break, as soon as my sister and I got back home and greeted everyone, ate, talk talk etc, we went to bed. And I can remember it so clearly baya.

When I put on my pajamas and readied myself for bed, I heard knocking on my door, so I opened it. To my surprise it was my little brothers, Andre and Don2x. They asked me, "can we sleep with you, Kuya?" hahahaha! So we shared the bed. It was so cute, 'te, kai my baby bro Andre carried a hug pillow that was 3/4 his height, Hahahahaha! Oh i can still remember when I used to put him on my chest to sleep when he was only 4-5 months old! And Don2x, i still can remember when I was 9 y/o and changed his diapers for the first time, hahahaha. I love my baby bros jud, the are like part of me. To hear their voice call out, "Kuya, Kuya" is like sweet nectar to the ears.
